Abstract:
:A series of unrelated steroids were tested for antimineralocorticoid activity by Kagawa's test in male rats and by modified test in female rats. Adrenalectomized male rats given 12 mcg desoxycorticosterone (DOC) and 2.5 ml normal saline received 1 mg of the test steroid, and their urine was analyzed for sodium and potassium. Results were expressed as the ratio (log sodium x 10)/potassium, and the end point was reversal of the DOC mineralocorticoid effect, i.e., increasing the ratio lowered by DOC. As expected, spironolactone and SC-11927 reversed DOC, but ethylestrenol, norbolethone, progesterone, triamcinolone, and prednisolone lowered the ratio as much as, or more than, DOC. In female rats given 10 mg of test steroids for 3 days, qualitatively similar results were obtained. These results showed that antimineralocorticoid activity is not associated with catatoxic (i.e., hepatic drug metabolizing) activity in spironolactone, norbolethone, progesterone, or ethylestrenol.
