Abstract:
BACKGROUND:Mistletoe therapy is frequently administered as a supportive treatment in diverse pediatric cancer entities including brain tumors. Medulloblastoma is the most common brain tumor in childhood. Its high risk to metastasize and its long-term sequelae caused by aggressive chemo- or radiotherapies are still challenging. MATERIAL AND METHODS:Effects of a lectin-rich mistletoe extract, abnobaVISCUM Fraxini, were investigated in two medulloblastoma cell lines (Daoy and ONS-76). Responsiveness of tumor cells was assessed by cell viability assays and xCELLigence real-time analyses. Moreover, impacts on proliferation, cell cycle and apoptosis were investigated. Apoptosis was studied by staining of vital mitochondria and assessing the involvement of caspases. In addition, effects on migration and invasion were analyzed. RESULTS:Both medulloblastoma cell lines were more susceptible to treatment with the mistletoe extract than a nontumorigenic fibroblast cell line. In mistletoe-sensitive Daoy cells, reduction of proliferation and induction of caspase-mediated apoptosis were observed upon administration of 0.05 and 0.5 mg/mL abnobaVISCUM Fraxini treatment, respectively. Furthermore, mistletoe extract inhibited migration and invasion properties in Daoy and significantly impaired invasive capabilities of ONS-76 cells. CONCLUSION:AbnobaVISCUM Fraxini has cell line dependent antitumoral effects in medulloblastoma models. These results call for further investigations, to reveal mechanistic insights into antitumorigenic properties of mistletoe extracts. :Hintergrund: Die Misteltherapie wird oft zur unterstützenden Behandlung verschiedener kindlicher Tumorentitäten wie dem Medulloblastom eingesetzt. Das Medulloblastom ist der häufigste bösartige Hirntumor bei Kindern. Das hohe Metastasierungsrisiko und die durch aggressive Chemo- und Radiotherapien verursachten Spätfolgen stellen Ärzte noch immer vor eine große Herausforderung. Material und Methoden: In dieser Studie wurden die Effekte des lektinreichen Mistelextraktes abnobaVISCUM Fraxini in zwei Medulloblastomzelllinien (Daoy und ONS-76) untersucht. Das Ansprechen der Zellen auf den Extrakt wurde in Zellviabilitäts-Assays (MTT) und in xCELLigence-Echtzeitanalysen erforscht. Außerdem wurden die Effekte auf die Proliferation, den Zellzyklus und die Apoptose ermittelt. Im Weiteren wurden Apoptosesignalwege durch das selektive Anfärben vitaler Mitochondrien und der Bestimmung des Einflusses von Initiator-Caspasen genauer untersucht. Zusätzlich wurden Versuche durchgeführt, um die Einflüsse des Mistelpräparats auf die Tumorzellmigration und die Invasion zu untersuchen. Ergebnisse: Unsere Untersuchungen konnten zeigen, dass die getesteten Medulloblastomzelllinien sensitiver waren als die verwendete nichttumorigene humane Fibroblasten-Zelllinie. Die sensitivste Zelllinie Daoy zeigte eine Verringerung der Zellproliferation und eine durch Caspase vermittelte Apoptose unter der Behandlung von 0,05 und 0,5 mg/mL abnobaVISCUM Fraxini, was mit einer Verringerung der mitochondrialen Integrität einherging. Zudem konnte eine Hemmung der Migrations- und der Invasionskapazität in Daoy-Zellen beobachtet werden. In ONS-76-Zellen konnte ein signifikanter Effekt auf die Invasion verzeichnet werden. Schlussfolgerung: Der Mistelextrakt abnobaVISCUM Fraxini hat antitumorale Effekte in Medulloblastomzellen, wobei das Ansprechen und die Effizienz abhängig von der Zelllinie zu sein schienen. Die Ergebnisse unterstreichen den Bedarf an weiteren Untersuchungen, die sich mit dem Wirkungsmechanismus von Mistelextrakten auf pädiatrische Tumorzellen befassen, um eine individualisierte Patientenbehandlung zu ermöglichen.
